Ardra profiles of bacteria and archaea in mangrove sediments with different levels of contamination in the estuarine complex of Paranaguá, Brazil BABT
Ribeiro,Catherine Gérikas; Steffens,Maria Berenice Reynaud; Etto,Rafael Mazer; Galvão,Carolina Weigert; Martins,César de Castro; Pedrosa,Fábio de Oliveira; Kolm,Hedda Elisabeth.
The mangrove's sediments from the coastal areas under human activities may contain significant contaminations by hydrocarbons, even when there are no visual evidences of it. The microorganisms are essential to these ecosystems, especially in the control of their chemical environment. Sediment samples were collected in two regions under different environment conditions (pristine and contaminated) of the Paranaguá Estuarine Complex (Paranaguá Bay and Laranjeiras Bay), Brazil. Aliphatic hydrocarbons were determined by the GC-FID to assess the status of contamination of the studied areas. The total DNA was extracted from these samples. The 16S rRNA gene was amplified by the PCR reactions with the pair of primers 21F and 958R for the archaeal domain, and 27F...

Assessment of viability in the bacterial standing stock of the Antarctic sea from the Indian side ArchiMer
Loka Bharathi, Pa; Nair, S; Bd De Souza, Mj; Chandramohan, D.
During the austral summer, we examined the bacterial population along the cruise track extending from 70degrees S and 18degrees E to 30degrees S and 35degrees E. During the cruise, three distinct fractions of the bacterioplankton viz. total count, total direct viable and retrievable counts were simultaneously enumerated in these waters. In the stations south of the convergent region designated as section I the population ranged within 10(8-9) L-1 whereas in the north of the region designated as section 11 they were one order higher and ranged from 10(9-10) L-1. The percentage of viability in the region was high corresponding to the generally high chlorophyll and primary productivity encountered in the eastern Aghulas bank. The study substantiates the...

Bacteria and protozoa populations in groundwater in landfill area in São Carlos, SP Rev. Microbiol.
Fusconi,Roberta; Godinho,Mirna Januária Leal.
The microbial populations of groundwaters were analyzed in a region under the influence of a landfill (piezometer L12) in the town of São Carlos, São Paulo, Brazil, and in an area not influenced by the landfill (piezometer L5). Heterotrophic bacteria were counted by spread plate method and the number of protozoa was estimated by the most probable number method. There was a larger number of organisms in well L12, with a mean value of 15.76 x 104 CFU/ml for bacteria and 9.7 MPN/ml for protozoa, whereas the mean values for piezometer L5 were 2.88 x 104 CFU/ml for bacteria and 3.4 MPN/ml for protozoa. The greater abundance detected in piezometer L12 may be related to the influence of the leachate through the landfill on the microbial populations, also...

Bacterial colonization of winter flounder Pseudopleuronectes americanus fed live feed enriched with three different commercial diets ArchiMer
Seychelles, L. H.; Audet, C.; Tremblay, R.; Lemarchand, K.; Pernet, Fabrice.
The proliferation of bacteria in intensive aquaculture systems may be responsible for poor growth and mass mortality of marine fish larvae. Essential fatty acids provided in the diet could protect larvae by modulation of the immune response via arachidonic acid (AA) and eicosapentaenoic acid (EPA). Winter flounder Pseudopleuronectes americanus larvae were fed rotifers Brachionus plicatilis enriched with three commercial diets containing different fatty acid profiles. Bacterial colonization on the gills and skin and in the intestinal lumen was evaluated at the end of the rotifer feeding period (day 26), and growth was surveyed until metamorphosis. At 26 days post hatching, larvae fed rotifers containing the higher AA content and with a higher...

Bacterial diversity in relatively pristine and anthropogenically-influenced mangrove ecosystems (Goa, India) BJM
Fernandes,Sheryl Oliveira; Kirchman,David L.; Michotey,Valérie D.; Bonin,Patricia C.; LokaBharathi,P.A..
To appreciate differences in benthic bacterial community composition at the relatively pristine Tuvem and the anthropogenically-influenced Divar mangrove ecosystems in Goa, India, parallel tag sequencing of the V6 region of 16S rDNA was carried out. We hypothesize that availability of extraneously-derived anthropogenic substrates could act as a stimulatant but not a deterrent to promote higher bacterial diversity at Divar. Our observations revealed that the phylum Proteobacteria was dominant at both locations comprising 43-46% of total tags. The Tuvem ecosystem was characterized by an abundance of members belonging to the class Deltaproteobacteria (21%), ~ 2100 phylotypes and 1561 operational taxonomic units (OTUs) sharing > 97% similarity. Bacterial diversity in soil in response to different plans, phosphate fertilizers and liming BJM
Silva,Paulo da; Nahas,Ely.
The diversity of bacterial isolates from soil in response to different plants (control, Brachiaria ruziziensis and Cajanus cajan), fertilization (control, simple superphosphate and rock phosphate) and liming (with and without lime) was evaluated. Phenotypic and physiological characteristics of the isolates were recorded and organized in a file to identify the bacteria. Among the isolates, 95% were Gram-positive and 5% Gram-negative rods. Soil cultivated with B. ruziziensis favored the nonsporing Gram-positive and Gram-negative rods compared to soils with C. cajan or uncultivated. Number of spore-forming Gram-positive rods were higher in plots with superphosphate than in unfertilized soil or soil fertilized with rock phosphate. In unfertilized plots, larger...

Bacterial Dynamics in a Microphytobenthic Biofilm: a Tidal Mesocosm Approach ArchiMer
Agogue, Helene; Mallet, Clarisse; Orvain, Francis; De Crignis, Margot; Mornet, Francoise; Dupuy, Christine.
In intertidal mudflats, during low tide exposure, microphytobenthos (MPB) migrate vertically through the surface sediment and form, with the heterotrophic bacteria, a transient biofilm. Inside this biofilm, multiple interactions exist between MPB and bacteria. These micro-organisms secrete a wide range of extracellular polymeric substances (EPS), which are major components of the biofilm matrix. In this study, we used a tidal mesocosm experiment in order to decipher the interactions of the MPB-EPS-bacteria complex within the biofilm. We tried to determine if the EPS could control bacterial activities and/or production and/or richness according to the age of the biofilm and to the immersion/emersion period. The dynamics of biomasses of MPB and prokaryotes,...

Elmnasser, Noura; Ritz, Magali; Leroi, Francoise; Orange, Nicole; Bakhrouf, Amina; Federighi, Michel.
Pulsed light is a new method intended for the decontamination of food surfaces using short, high frequency pulses of an intense broad spectrum. The effects of broad spectrum pulsed light on the survival of Listeria monocytogenes Scott A, Listeria monocytogenes CNL, Pseudomonas fluorescens MF37 and Photobacterium phosphoreum SF680 populations on agar and in a liquid medium were investigated during this study. The sterilisation system generated 1.5 J cm(-2) per pulse with eight lamps for 300 mu s. In the case of surface-seeded cells, a 7.8, 8.14 and > 7.14 log reduction was obtained for L. monocytogenes, Ps. fluorescens MF37 and Ph. phosphoreum, respectively, after a single pulse of treatment. Inactivation levels were lower for depth-plated cells: indeed,...

Bacteries associees aux production de Brachionus plicatilis ArchiMer
Nicolas, Jean-louis; Joubert, M.
Bacteria associated with semi-continuous culture of Brachionus plicatilis , which is used as the first food of marine fish larvae, were counted on Zobell 2216 E medium and identified. The numbers of bacteria were very high in both Brachionus (about 10 super(11) bact. per ml of digestive content) and in water (from 1.6.10 super(6) to 4.7.10 super(7) bact. per ml). The cultures were studied at driving a time when mortalities occurred frequently. The introduction of a Pseudomonas) phenon at a proportion of about 50% to the dominant microflora persisted in time and was little influenced by diet. It has been the cause of the repression of the Vibrio, Aeromonas group which was found in an unusually small proportion. It induced a lack of microflora diversity and...

Bacterial infections occurred during the development of marine fish farming of species such as turbot, sea-bass, sea-bream or salmon and trout. The main-specific bacteria involved in these diseases observed belong to 3 groups: the Vibrionaceae for Vibrio anguillarum (type 408) and Aeromonas salmonicida , the causative agents of respectively vibriosis and furinculosis; the Corynebacteriaceae for Renibacterium salmoninarum , responsible for bacterial kidney disease (BKD), the Enterobacteriaceae for Yersinia ruckeri , the causative agent of enteric redmouth disease. The clinical signs typical of those diseases always depend on the rearing and environmental conditions. The losses resulting from these bacterial diseases and the persistence of the bacteria in...

Bacterioplancton in different tides of the Perequê tidal creek, Pontal do Sul, Paraná, Brazil BJM
Kolm,Hedda Elisabeth; Andretta,Lucimar.
The influence of tidal phase on bacteria abundance in surface waters was studied in five stations along the Perequê tidal creek (Paraná State, Brazil), at spring and neap low and high tides in July 1999. The stations were located from the mouth (St.1) to the creek's head (St. 5). The following parameters were analyzed: salinity, temperature, pH, dissolved oxygen, total carbon dioxide, seston, particulate organic matter, pluviosity, chlorophyll "a", total heterotrophic bacteria, bacterial biomass, cultivable heterotrophic aerobic halophilic and halophobic bacteria, total coliforms and Escherichia coli. The results showed that the highest values of salinity, pH and dissolved oxygen at stations 1 to 4 were registered during the spring high tide. The amount of...

Bacterivory by benthic organisms in sediment : Quantification using 15N-enriched bacteria ArchiMer
Pascal, P.y.; Dupuy, C; Mallet, C; Richard, P; Niquil, N.
The fate of benthic bacterial biomass in benthic food webs is a topic of major importance but poorly described. This paper describes an alternative method for evaluation of bacterial grazing rate by meiofauna and macrofauna using bacteria pre-enriched with stable isotopes. Natural bacteria from the sediment of an intertidal mudflat were cultured in a liquid medium enriched with 15NH4Cl. Cultured bacteria contained 2.9% of 15N and were enriched sufficiently to be used as tracers during grazing experiments. Cultured bacteria presented a biovolume (0.21 μm3) and a percentage of actively respiring bacteria (10%) similar to those found in natural communities. The number of Operational Taxon Units found in cultures fluctuated between 56 and 75% of that...

Bacterivory in the common foraminifer Ammonia tepida: Isotope tracer experiment and the controlling factors ArchiMer
Pascal, P.y.; Dupuy, C; Richard, P; Niquil, N.
The majority of sediment dweller foraminifera are deposit feeders. They use their pseudopodia to gather sediment with associated algae, organic detritus and bacteria. Uptake of bacteria by foraminifera have been observed but rarely quantified. We measured uptake of bacteria by the common foraminifera Ammonia tepida using 15N pre-enriched bacteria as tracers. In intertidal flats, seasonal, tidal and circadian cycles induce strong variations in environmental parameters. Grazing experiments were performed in order to measure effects of abiotic (temperature, salinity and irradiance) and biotic (bacterial and algal abundances) factors on uptake rates of bacteria. In mean conditions, A. tepida grazed 78 pgC ind-1 h-1 during the first eight hours of incubation,...
